Ligustilide is recognized for its broad pharmacological potential, often attributed to its ability to modulate multiple biological pathways simultaneously.
1Neuroprotection: It crosses the blood-brain barrier and has shown potential in treating brain injuries and neurodegenerative diseases like Alzheimer's by reducing neuroinflammation and oxidative stress.
Anti-inflammatory & Antioxidant: It inhibits pro-inflammatory cytokines (like TNF-α and IL-6) and activates the Nrf2/HO-1 defense pathway to combat oxidative damage.
Gastrointestinal Health: Recent studies suggest it may treat GI disorders by targeting proteins like PTGS2 (COX-2) and EGFR to reduce inflammation and promote healing.
Anticancer: It has demonstrated the ability to inhibit tumor growth, induce apoptosis (cell death), and overcome chemoresistance in various cancers, including gastric, breast, and bile duct cancer.
Cardiovascular Effects: It acts as a vasodilator by inhibiting calcium influx in vascular smooth muscle, which may help manage hypertension and atherosclerosis.

Note on Stability: Ligustilide is highly unstable and sensitive to light, heat, and oxygen, which leads to rapid degradation and low oral bioavailability (approximately 2.6% in rats). Researchers are exploring more stable crystalline derivatives and nano-delivery systems (like liposomes or nanoemulsions) to improve its therapeutic effectiveness.
